Amifostine is a broad-spectrum cell protectant. It was once used as a military anti-radiation agent, and is now used as a protective drug for radiotherapy and chemotherapy. It was effective in reducing the side effects of anti-cancer drugs such as cyclophosphamide, chloroplatin, doxorubicin and anti-HIV drugs such as 3-azido-3-deoxythymidine, and Chemicalbook did not affect the efficacy of anti-cancer drugs or anti-HIV drugs. Amifotin is currently available as a freeze-dried powder injection from Life Sciences Inc., under the trade name Ethyol. The product is available only as a lyophilized powder injection, but the amorphous form produced by the low pressure lyophilized process is heat unstable. Amifostine trihydrate (WR2721 trihydrate) is a broad-spectrum cytoprotective agent and a radioprotector. Amifostine trihydrate selectively protects normal tissues from damage caused by radiation and chemotherapy. Amifostine trihydrate is potent hypoxia-inducible factor-α1 (HIF-α1) and p53 inducer. Amifostine trihydrate protects cells from damage by scavenging oxygen-derived free radicals. Amifostine trihydrate reduces renal toxicity and has antiangiogenic action.
