Tropifexor, also known as LJN452, is a farnesin X receptor agonist for the treatment of cholestatic liver disease and non-alcoholic steatohepatitis (NASH). By measuring the induction of FXR target genes in various Chemicalbook species, in vivo activity was demonstrated in a rodent PD model. Tropifexor has entered phase 2 human clinical trials in NASH and PBC patients. Tropifexor(LJN452) is a novel and effective FXR agonist with an EC50 of 0.2nM in HTRF experiments. Chemicalbook showed no significant off-target effects against other enzymes, ion channels, nuclear receptors, and GPCR, and was more than 10,000 times more selective for FXR than for other targets.
