BLU9931 is a potent, selective, and irreversible FGFR4 inhibitor with an IC50 of 3 nM and a selectivity of 297,184 and 50 times higher than that for FGFR1/2/3, respectively. In M-DA-MB-453 cells, BLU9931 effectively inhibited the phosphorylation of FGFR4 signaling pathway. Blu9931 inhibited the proliferation of HCC cell lines, such as Hep3B, Huh-7, Chemicalbook, and JHH-7, expressing an intact FGFR4 signaling complex with EC50<1 μm. BLU9931 also inhibits proliferation in PDX-derived cell lines with an intact FGFR4 signaling pathway. In mice loaded with FGF19-amplified Hep3B liver tumors, BLU9931(300mg/kg, P.O.) caused tumor degeneration and prevented tumor-induced weight loss. Blu9931 (300mg/kg, P.O.) treatment also resulted in tumor degeneration in Chemicalbook loaded mice with FGF19- overexpression of PDX-derived LIXC012 xenograft.
